Some basic comfort with NVDA helps, but it's not required. Each module walks through the component before asking you to test it, so you'll pick up the keystrokes and workflow as you go. If you want a guided intro first, my free NVDA resources cover installation and the basics.
WCAG Quest is a short, playable introduction to WCAG concepts - one sitting, gamified. Hands-On, Sight Off is a much larger, self-paced practice course covering over 20 real interactive component patterns in depth. Many people start with WCAG Quest, then move here for the deeper practice.
It's self-paced with no deadline, so you can work through all the modules in one sitting or spread them across several sessions - most learners spend a few hours total, depending on how much time they spend testing each pattern.
